The Synapt G2-S is a Q-TOF-based mass spectrometer with an additional ion mobility cell,
allowing for orthogonal separation of ions based on their shape and conformation.
Equipped with an intermediate quadrupole (8k), this highly versatile instrument can be used not only for peptide identification
(LC-MSE data-independent acquisition) but also for denaturation and native MS analysis.
The Waters nanoACQUITY UPLCTM system is designed for separations from capillary to nanoscale.
Its sensitivity, resolution, and reproducibility are well-suited for biomarker discovery
and proteomics applications, including protein identification and characterization.

HPLC Waters Binary Solvent Manager:
The nanoACQUITY binary solvent manager is a high-pressure pump that moves solvents through the system.
It ensures a smooth (pulsation-free) solvent flow at flow rates of 0.20 to 5.00 l/min
(in closed loop) or 100.0 l/min (in open loop) at 69,000 kPa (690 bar, 10,000 psi).
The solvent manager can generate high-pressure gradients with minimal gradient delay.
Operation of the binary solvent manager
Each of the two independent pump systems of the solvent manager, Pump A (left) and Pump B (right),
contains two linear drives.
Each left and right actuator pair consists of a single reciprocating serial pump,
which delivers a precise flow of a single solvent.
The two pump systems combine their two solvents at a mixing T-piece.
From there, the solvent mixture flows to the sample manager.
To create gradients and mixtures, the chromatography software controls the mixing ratio of the two solvents,
by varying the flow of Pump A relative to that of Pump B.
A pressure transducer in each pump head transmits pressure data to the solvent manager,
whose firmware measures the pump head pressure during the pumping cycle.
This allows the solvent manager to adjust the pre-compression,
to ensure a consistent solvent supply
and minimize pump-induced detector baseline disturbances.
Flow control modules
The flow control modules enable the binary solvent manager to perform nano-flow chromatography
by monitoring the solvent flow in each solvent channel.
The flow control is calibrated for water, acetonitrile, and methanol.
The two mass flow sensors of the flow control modules measure the flow from each pump separately,
before they are mixed.
The binary solvent manager monitors the output and adjusts the solvent flow accordingly,
to ensure a precise flow of 0.20 to 5.00 µl/min.
Solvents are mixed at the T-piece outlet of the flow control module.
Auxiliary solvent manager
The nanoACQUITY auxiliary solvent manager contains two isocratic high-pressure pumps,
which move solvents through the system and ensure a smooth (pulsation-free) solvent flow.
Pump A is used for sample loading.
• Pump B is used for adding the NanoLockSpray-Lock mass, for trapping with two pumps, and for 2D online separation.
In the NanoLockSpray-Lock mass addition mode, the pump operates at flow rates of 0.10 to 5.00 µl/min.
Flow control module
The flow control module enables the auxiliary solvent manager to perform nano-flow chromatography
by monitoring the solvent flow in the solvent channel.
The flow control is calibrated for water, acetonitrile, and methanol.
The mass flow sensor of the flow control module measures the flow from the pump.
The auxiliary solvent manager monitors the output and adjusts the solvent flow accordingly,
to achieve a precise flow of 0.20 to 5.00 µl/min.
Sample manager
The sample manager injects the samples it takes from microtiter plates or vials into the chromatographic flow stream.
In maximum throughput mode, the sample manager can perform an injection in about 45 seconds,
including a single standard wash, or it can do this in less than 60 seconds,
including a double standard wash.
The sample manager accepts plates with standard print, 5.03 $0.02 inches x 3.365 $0.02 inches,
which comply with SBS/ANSI standards (maximum height of 2.2 inches, including covers).
The sample manager can store samples at any temperature between 4 and 40 °C (39 and 104 °F)
under ambient conditions of 25 °C (77 °F) or less.
Heating and trapping module
The heating and trapping module is attached to the sample manager and serves as its upper cover.
The column base of the heating and trapping module can accommodate a nano T-piece and any analytical Waters column
with an inner diameter of up to 300 micrometers and a length of 250 mm.
The column space heats to temperatures of 5 °C (9 °F) above ambient temperature up to 65 °C (149 °F).
To reduce dispersion associated with dead volume and minimize the length of tubing between system modules,
the column base can be pivoted outward to any position between 0° and 180°.
In the 0° "Home" position, the column base is directly above the sample manager
and can be connected to an optional optical detector.
In the 180° "away" position, the analytical column can be connected to a mass spectrometer
(on the right side of the system).
